Cloning and
classification of bt endotoxin gene from Bacillus Thuringiensis
Thai isolates of JC276, JC281 and JC291, that test for regulate corn borer
worms, in Thailand. Two pair of
Universal oligonucleotide primers were designed to amplified PCR Cry-type gene
fragment of the DNA template from B.thuringiensis. The amplified Cry gene products of all
three isolate are 3 kilobase (K5Un3 and K3Un3 primers give 1.6 kilobase
fragments and K5Un2 and K3Un2 primers give 1.4 kilobase fragments). The restriction fragment length
polymorphism (RFLP) pattern of PCR-amplified fragment revealed 3 possible
distinct Cry-type genes included CryIA(a), CryIB and possible CryID or CryI(b)
or Cry IF. Among these the
sequence of those genes were found to be different from the published Cry gene
sequence. Interestingly, Cry-type
gene detect from B. thuringensis JC276 and JC281 were distinct
from Cry-type gene from JC291.
However, all Cry-type genes isolate from Thai isolates are differed in
gene sequencing from reported Cry-type gene in database.
